RESUMO

The experiment evaluated the effect of adding cholesterol-loaded cyclodextrin (CLC) to Prochilodus lineatus fish (Curimata) semen on post-thaw sperm quality. Twelve adult fish were used for sperm collection after induced spermiation with carp pituitary gland. The semen was diluted and treated with CLC in concentrations of 0 (control), 0.5, 1.0, 2.0, 3.0, and 4.0 mg for 120 × 106 spermatozoa/ml, loaded in 0.5 ml straws, packaged and placed in dry vapor vessel cylinders for 24 h before being submerged in liquid nitrogen for storage. The samples were thawed in a water bath at 60 °C for 8 s, and the sperm parameters evaluated were motility, activation duration, longevity, plasma membrane integrity, and morphology. Data were tested for normal distribution and ANOVA, followed by Friedman test (P < 0.05). Spermatozoa treated with CLC displayed higher motility than the control (P < 0.05). The duration of sperm activation was longer in sperm treated with 0.5, 1.0, and 2.0 mg of CLC than in control (P < 0.05). The membrane integrity was higher in sperm treated with 0.5, 1.0, 2.0, and 3.0 mg of CLC than in control and four mg-treated samples (P < 0.05). The sperm longevity and morphology alterations did not differ between treatments (P > 0.05). Adding 0.5, 1.0, or 2.0 mg of CLC in Prochilodus lineatus semen before cryopreservation improves sperm motility and membrane integrity.

RESUMO

Abstract In order to evaluate aspects of reproductive biology of the "pequira" ( Bryconamericus stramineus) in the elevator of the Funil Dam - MG, fish capture was carried out from November 2008 to January 2009 and 317 individuals were collected. The mean standard length (SL) of the population was 4.96 cm and mean weight 1.80 g. The females had SL of 5.0 cm, while males had a SL of 4.6 cm. A sex ratio of 2.20: 1 (females: male) was observed. Our results show that 73% of the individuals analyzed were considered adults. The species presented low fecundity, mean of 470.9 oocytes per female and a mean diameter of 221.08 μm, with an increase in oocyte diameter over the evaluation period. The length of the first gonadal maturation was estimated at 5.0 cm. The results obtained in this work suggest that the reproductive cycle of the species occurs in the analyzed period. Although this species does not have migratory reproductive habits, the presence of adults in the reproductive stage was observed in the transposition area, which suggests a search for new environments for spawning.

BACKGROUND: Semen cryopreservation is a technique widely used in commercial and conservation fish farms. However, the success of the technique will depend on the diluent and cryoprotectant solutions added to the semen prior to the freezing procedure. OBJECTIVE: To evaluate the effects of different doses of melatonin on motility, morphology and fertilization capacity of cryopreserved semen of Prochilodus lineatus. MATERIALS AND METHODS: Semen was diluted in cryoprotectant solution with added melatonin, resulting in three treatments. A Control Solution (DMSO8% + BTS5%), was compared with T1, T2 and T3 treatments: BTS (5%) + DMSO (8%) plus concentrations of 1, 2 and 3 mM of melatonin, respectively. The diluted semen was frozen in a cylinder of vapour nitrogen and stored at -190°C. The straws were thawed in a water bath at 40°C for 12 s for analysis of sperm motility and morphology, and for evaluation of fertilization rate. The data were submitted to ANOVA, and the means were compared by the Tukey test. RESULTS: There was no statistical difference (P>0.05) between the progressive motility rates and the velocities of the cryopreserved semen in the different treatments. The total motility rate was higher (P <0.05) in the groups T1 and T2 when compared to T3, but did not differ from control. All treatments had similar values of total anomalies after thawing and did not differ in fertilization capacity (P>0.05). CONCLUSION: The addition of melatonin on the cryoprotectant solution did not promote changes on motility, morphology and fertilization capacity of cryopreserved semen of Prochilodus lineatus.

RESUMO

OBJECTIVE: To identify which sperm characteristics were able to predict more accurately the quality of curimba (Prochilodus lineatus) semen upon freezing using canonical correlation analysis. METHODS: Eleven fish breeders with initial mean weight of 705.21 ± 111 g were used. For cryopreservation, 200 µL of semen were taken from each animal and diluted in the cryoprotectant solution (10% dimethyl sulfoxide and 5% Beltsville Thawing Solution Minitub) in a 1:4 ratio and placed into 0.5-mL straws. Sperm characteristics (motility, sperm abnormalities, total antioxidant activity and lipid peroxidation) were evaluated. A randomized block design with duplicate samples per treatment (fresh and frozen semen) was used. The block factor was the animals, and the experimental unit the ejaculates. Canonical correlation was used to evaluate the association between sperm characteristics of fresh semen and thawed semen. RESULT: There was a significant association (P = 0.10) among the variables measured in fresh semen with the variables measured in thawed semen, and 78.6% of the difference observed in the thawed semen can be attributed to variation of variables measured in fresh semen. Sperm motility, motility duration and antioxidant activity of the thawed semen showed an inverse relationship with those of the fresh semen; whereas the minor sperm abnormalities, major sperm abnormalities and lipid peroxidation showed a direct relationship with those of the fresh semen. Only the rate and motility duration of the thawed semen presented high correlation (-0.63 and -0.73, respectively) with the canonical variable represented by the sperm characteristics of fresh semen. CONCLUSIONS: The rate and motility duration of fresh semen may be used to predict the quality of the thawed sperm in Prochilodus lineatus.

RESUMO

BACKGROUND: Cryopreserved semen could facilitate procedures during the artificial reproduction in fish. Factors affecting cryopreservation efficiency are important to define efficient protocols. OBJECTIVE: This study investigated the application of cryoprotectants on the quality of piracanjuba fish semen, the sperm concentration required for oocyte fertilization and spermatic activation. MATERIALS AND METHODS: We evaluated two intracellular cryoprotectant solutions (DMSO and methanol) and two extracellular cryoprotectant solutions (egg yolk and lactose) to cryopreserved piracanjuba semen. Sperm motility rate, motility duration and spermatic alterations were assessed. CONCLUSION: The protocol for piracanjuba semen cryopreservation can use solutions including either DMSO or methanol as intracellular cryoprotectant and egg yolk or lactose as extracellular cryoprotectants.

RESUMO

This study evaluated the reproductive dynamics and gonadal morphology of piaparas (Leporinus obtusidens) captured downstream from the Funil Hydroelectric Power Station. The 107 piaparas evaluated were captured monthly from Sep 2006 to Aug 2007. The data were stratified among the seasons to evaluate gonad maturation and the reproductive period was determined by assessing the gonadosomatic index, hepatosomatic index, abdominal fat index (AFI), weight:length ratio, condition factor and sexual proportion. The reproductive period lasted from winter to summer (Jul-Mar) in females and from spring to autumn (Sept-Jun) in males. AFI was high during and after the reproductive period in females and only during the reproductive period in males. The highest HIS values for males and females were obtained in spring and summer (Sept-Mar). The females observed did not spawn and showed follicular atresia. These characteristics suggest that L. obtusidens does not use the study area as a reproductive site and probably migrates for this purpose.

The objective of this study was to evaluate the influence of different photoperiods on physiological parameters related to stress in Nile tilapia fingerlings (Oreochromis niloticus). A completely randomized design with five treatments (0L, 6L, 12L, 18L, 24L) and four replications was used, and the aquarium as the experimental unit. One hundred and sixty tilapia fingerlings, 3.21±0.05g and 4.35±0.07cm were distributed in 20 aquariums of 20L in a recirculation system with temperature controlled at 27 º C and density of eight fish per aquarium. Feeding was performed twice daily with extruded feed containing 40% crude protein, for a period of 75 days. At the end of experimental period, the data of survival, cortisol, glucose and hematocrit were subjected to analysis of variance and, subsequently, in case of significance (P<0.05) a Tukey test 5% was performed to compare means. Manipulation of photoperiod shows no alteration in the homeostasis of juveniles, since there is no difference in survival, blood glucose, hematocrit and plasma cortisol (P>0.05).

RESUMO

The objective of this study was to evaluate the addition of IGF-I to pig insemination doses stored at 15°C, in conjunction with the addition of different amounts of vitamin E (α-tocopherol). Semen samples (n = 12) from four boars were treated by the addition of different concentrations of vitamin E, ranging up to 400 µg/ml. Immediately after processing and after the doses had been stored at 15°C for 24 or 72 h, samples were warmed at 37°C and 30 ng/ml of IGF-I was added. The assessments were made after 10 and 120 min of IGF-I addition. There was a minor effect of the vitamin E added before cooling and IGF-I added after storage on sperm quality. The addition of 400 µg/ml of vitamin E to diluted semen reduced (P < 0.01) the malondialdehyde (MDA) production in boar semen stored at 15°C for 72 h, regardless of the addition of IGF-I as additive during a 120 min incubation period at 37°C. In these conditions, IGF-I also reduced (P < 0.05) the MDA production in semen samples without addition of vitamin E. IGF-I in the presence of vitamin E reduced (P = 0.03) the glucose intake in freshly diluted boar semen samples before cooling. It was concluded that the addition of 400 µg/ml of vitamin E reduces the MDA production in boar semen stored at 15°C for 72 h, regardless of the presence of IGF-I additive. The addition of IGF-I in doses stored for 72 h with vitamin E ensures higher sperm motility after 120 min of incubation at 37°C.

This study aimed to evaluate the influence of cryoprotectants, thawing times and temperatures on pacu (Piaractus mesopotamicus) sperm abnormalities. Semen samples from four pacu were collected and diluted in 2 solutions, cryoprotectant dimethylsulfoxide (DMSO) 10% and methanol 10%. After thawing, semen was collected for morphological analysis, evaluating primary and secondary abnormalities. There was no interaction (P>0.05) of secondary abnormalities, abnormalities in total and normal sperm in the different treatments. The primary abnormalities showed interaction between temperature and thawing time, where the higher temperature was associated with the shortest time (60°C for 8s) or the lowest temperature associated with longer thawing (40°C for 12s) provided under (P<0.05) percentages of spermatozoa with primary abnormalities. In the process of semen cryopreservation of P. mesopotamicus using the cryoprotectants DMSO and methanol, it is recommended that thawing be performed at 40° C for 12 seconds or 60°C for 8 seconds.

RESUMO

The objective was to evaluate the influence of the timing of hormonal induction, using gonadorelin or common carp pituitary extract (CPE), on the reproductive activity of female Astyanax bimaculatus. Fish (N = 44) were weighed, measured, and acclimatized to experimental conditions with a photoperiod of 12 h:12 h light:dark (L:D) for 10 days. Ovulation was induced with a single dose of CPE (6 mg/kg) or gonadorelin (80 µg/kg), given at 12:00 (halfway through the light phase (LP) or 24:00 (halfway through the dark phase (DP), in a 2 × 2 factorial design. The time of ovulation was calculated in degree hours and daily motor activity was recorded using a photocell. The fish were killed and the liver and gonads were weighed for calculation of gonadosomatic (GSI) and hepatosomatic (HSI) indexes, respectively. Absolute fecundity (AF), absolute fecundity relative to weight (AFRW) and length (AFRL), diameter of oocytes (mM), and percentage of oocytes with the germinal vesicle in a peripheral position (PPGV) were recorded. All females responded (ovulated). The female Astyanax bimaculatus had twilight motor activity rhythm. Females given CPE at 12:00 had a higher (P < 0.05) percentage of oocytes with the germinal vesicle in a peripheral position compared with the group that received gonadorelin in the same period (95 ± 6 vs. 79 ± 21%, mean ± SD). The absolute fecundity relative to weight was higher in groups induced at 12:00, regardless of the hormone used (LP: 805 ± 448 and 700 ± 214, for CPE and gonadorelin, respectively; dark phase: 580 ± 396 and 529 ± 105, P < 0.05). Both times used for hormonal induction with CPE and gonadorelin were suitable for inducing reproduction in lambari, although induction with CPE in LP had the best results.

The objectives of the present study were to evaluate the legal prescription adequacy in one drugstore located in the south of Minas Gerais state, in order to establish comparisons between the prescriptions of public and private services and to detect the most commonly classes of medicines prescribed. It was analyzed 960 prescriptions in accordance with the current law. The data were analyzed by descriptive statistics and compared using frequency distribution test(Chi-square test) considering private and public sources of prescription as the dependent variable. Considering special control prescriptions, the extensive description of the amount of medicine was fulfilled more frequently by the private sector. However, for the notifications; legibility, dosage, pharmaceutical form, date of issue and presence of stamp were more frequently present in the public service, while assignment of the Federative Union, the extensive description of the amount of medicine and the dosage was more present in the private sector. None of the evaluated prescriptions was considered totally adjusted with law 344/98. The prescribed medicines more commonly found were benzodiazepines for prescription notifications and antidepressants in special control prescriptions. It was concluded that the medicine prescriptions subjected to special control are frequently not adequate to the current law.

RESUMO

OBJECTIVE: To evaluate in vitro IGF-I treatment during warming of storage-cooled boar semen and its effect on seminal quality parameters and metabolism in spermatic cells. DESIGN: Semen samples (n=7) warmed after stored at 15°C for 24 or 72h were divided into four equal parts. Different IGF-I concentrations (0, 50, 100 and 150ng/mL) were added to the semen samples. The samples were incubated at 37°C, and assessments were made after 0 and 120min of incubation. RESULTS: For semen samples that were stored for 24h, the addition of IGF-I had no effect (p>0.05) on the total motility and intensity of movements by spermatic cells, osmotic resistance, live:dead cell ratio or total spermatic abnormalities. However, incubation with 150ng/mL IGF-I did decrease glutathione peroxidase activity (p<0.05) and reduce lipid peroxidation after 120min of incubation. For semen samples stored for 72h and incubated with IGF-I for 120min, there was a linear relationship between the IGF-I concentration and the live:dead ratio (p<0.05). There was a quadratic relationship between the IGF-I concentration and both the osmotic resistance (peak results at IGF-I=62.4ng/mL) and glutathione peroxidase activity (peak results at IGF-I=77.8ng/mL). There was no effect on lipid peroxidation (p>0.05) after 120min of incubation. Addition of IGF-I also decreased fructose utilization by spermatic cells regardless of semen storage time (p<0.05). CONCLUSION: This study suggests that IGF-I may be beneficial to semen stored for longer periods of time. Adding 150ng/mL IGF-I improved the quality of semen stored for 24h, and adding 78ng/mL IGF-I improved the quality of semen stored for 72h.

RESUMO

This study investigated the application of intra- and extra-cellular cryoprotectant combinations on the quality of curimba Prochilodus lineatus semen subjected to cryopreservation. Semen treatments were tested with 8% DMSO or methanol as intracellular cryoprotectant, 5% egg yolk or lactose as extracellular cryoprotectant and 5% BTS. These cryoprotectant combinations are suitable for curimba but have not been tested at the lesser concentrations proposed or in combination with BTS. Semen samples collected from 19 curimbas were diluted into one of four cryoprotectant combinations: DMSO+yolk; DMSO+lactose; methanol+yolk; and methanol+lactose. After dilution, semen samples were cryopreserved in 0.5 mL straws for 10 days in a liquid nitrogen tank. Semen was thawed in a water bath at 60°C for 8s. We evaluated the quality of fresh, diluted (pre-freezing) and post-freezing semen according to sperm motility rate (%) and duration (s). Sperm morphology was also analyzed in thawed semen. Sperm motility rate decreased progressively after dilution and thawing. The motility rate in post-freezing semen was higher in the treatments using DMSO+lactose and methanol+yolk. Sperm motility duration in post-freezing sperm was greater in the treatments using methanol rather than DMSO as intracellular cryoprotectant, irrespective of the extracellular cryoprotectant used. Abnormality frequency in thawed sperm was less in semen treated with egg yolk than with lactose. Thus the use of methanol intracellular cryoprotectant is recommended along with yolk extracellular cryoprotectant in the cryopreservation process for curimba semen.

The addition of oil sources and dietary vitamin E supplementation was evaluated on the characteristics of fresh boar sperm. Twenty-four mature Dalboar 85 boars of proven fertility and in routine semen production for artificial insemination were randomly distributed in a factorial arrangement 2 X 3, with two oil sources, soy and salmon, and three antioxidant levels: 150, 300, and 450mg/kg of vitamin E. Volume, total motile sperm, hyposmotic swelling test, percentage of live cell, and morphology did not differ (P<0.05) between the animals from the treatments. Salmon oil improved (P<0.05) sperm vigor. The vitamin E added to the diet improved (P<0.05) the sperm concentration, and there was no difference between the oil sources (P>0.05). Salmon oil treatment showed the lowest (P<0.05) concentration of total antioxidants in the semen. Vitamin E and concentration of total antioxidant showed a linear effect (P<0.05). The semen vigor and the concentration of total antioxidants were improved by oil sources, but the sperm concentration was dependent on the levels of vitamin E. Therefore, salmon oil can be used in the diet of male pigs.

The effects of ascorbate on the haematocrit and blood glucose level were evaluated in Nile tilapia alevins (Oreochromis niloticus) submitted to a transport simulation. Three experimental diets with different levels of vitamin C (16, 500 and 1000mg/kg) were given for 14 days before the simulation of the transport. The treatment containing 16mg of vitamin C showed the highest level of glucose after the simulation (108.5mg/dl immediately after the transport and 91mg/dl 12 hours after the transport). The vitamin C concentration of 1000mg/kg was the most efficient treatment to control glycemia increases (94.6mg/dl immediately after the simulation and 74.4mg/dl 12 hour after simulation). In the 500mg/kg treatment, the glucose level was 91.4mg/dl immediately after the simulation and 103.8mg/dl 12 hours after the simulation. The haematocrit values did not show any significative variation (P<0.05). The supplementation with 1000mg/kg of vitamin C for a 14 days period can be used in a prophylactic way to soften glycemia increases in Nile tilapia alevins submitted to transport stress

With the objective of evaluating the effects of different photoperiods on the development and testicular histology of light roosters, an experiment was conducted in the Animal Science Department of the Federal University of Lavras. 48 roosters of the Lohman LSL strain, aged 133 days were utilized, they themselves being submitted to 3 treatments: 1) Continuous photoperiod, 2) Intermittent photoperiod and 3) Growing natural photoperiod. The animals were allotted to a completely randomized experimental design, with 4 replicates being the experimental unit being made up of 1 animal. The experimental period lasted 140 days and the variables evaluated were: body weight, weights of the right (PTD) and left testes (PTE), in grams; diameter of the seminiferous tubules (DTS) and thickness of the seminiferous epithelium (EES), in micrometers and number of cells of the seminiferous tubules (Sertoli cells, rounded spermatogonia and spermatids). The differences between treatments were compared by the Scott Knott test (5%). A significant effect was not observed (P>0.05) of the photoperiods and the ages on the analyzed variable, with exception of the number of spermatogonia that had presented quadratic variation with the increase of the age of the animals. It follows that photoperiods continuous, intermittent or growing natural light can be utilized for creation of light roosters over the reproductive period without harming the testicular parameters. Had to the lesser expense of energy that can be gotten with the use of Growing natural photoperiod, this would be the recommended program more for themales, with regard to the testicular characteristics

The experiment was conducted in the Furnas hydrobiology and picisculture station situated in São José da Barra - Minas Gerais in Brasil with a view to verifying the best ratio between males and females and also ideal dose of 17-∝-methyltestosterone, adjusting a management methodology in the reproduction of Nile tilapia fish. Two experiments were conducted, the first one verifying the best ratio between males and females (1:1; 1:1.4; 1:2; 1:3; 1:5) and the parameters evaluated were: eggs production; spawning number and hatching percentage. In the second experiment was observed the ideal dose of the hormone 17-∝ methyltestosterone, its being made up of five hormone level: treatment A with 0 mg of hormone/kg of ration, treatment B with 30 mg de hormone/kg of ration, treatment C with 60 mg of hormone/kg of ration, treatment D with 90 mg of hormone/kg of ration and treatment E with 120 mg of hormone/kg of ration and the parameters evaluated were: percentage of male without a hormonal treatment and percentage of males after hormonal treatment. In the first experiment, the ratio of one male to one female produced the highest number of eggs (P<0.05). In the second experiment there were significant differences (P<0.01) for dosage of 17-α-metiltestosterona equal or superior to 37.5 mg of hormone/kg of ration, which produced 100% of males (AU)
